Biological Background: WNT2 Function and Localization
- Protein Wnt-2 (also known as Int-1-related protein, IRP) is a member of the Wnt family of secreted signaling proteins that play key roles in development and tissue homeostasis.
- Functions as a ligand for frizzled family receptors and activates the canonical Wnt/β-catenin signaling pathway, leading to TCF/LEF-dependent transcription. (PubMed:20018874) Related references: PMID:20018874
- Acts as an upstream regulator of FGF10 expression and is essential for embryonic lung branching morphogenesis.
- Contributes to embryonic brain development by regulating the proliferation of dopaminergic neuronal precursors and neurons. (By similarity)
- Localizes to the extracellular space and extracellular matrix, consistent with its role as a secreted morphogen.
- Expressed in the thalamus of the brain, in fetal and adult lung, and in placenta.
- Contains conserved cysteine residues forming disulfide bonds, and undergoes N-glycosylation and palmitoylation, which are important for its secretion and receptor interaction.
- Keywords: Developmental protein, Disulfide bond, Extracellular matrix, Glycoprotein, Lipoprotein, Secreted, Signal, Wnt signaling pathway.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the mature human WNT2 protein (aa 26–360), so the antibody is expected to recognize the secreted form. Polyclonal nature may provide detection of multiple epitopes.
- For Western blotting (WB), the predicted molecular weight is ~40 kDa for the unmodified protein; however, post-translational modifications like glycosylation may result in a higher apparent molecular weight. Use appropriate reducing conditions.
- For immunohistochemistry (IHC-P), antigen retrieval (e.g., citrate buffer, pH 6.0) may improve epitope exposure. Optimize dilution and detection system for your specific tissue type.
- In ELISA, this antibody can be used as a capture or detection reagent for WNT2; validate with recombinant protein or known positive samples.
- Cross-reactivity with mouse and rat WNT2 has been reported, but sequence homology should be considered. Validate in your model system.
