Biological Background: WNK3 Function and Localization
- WNK3 (Protein kinase lysine-deficient 3) is a serine/threonine-protein kinase encoded by the WNK3 gene (also known as KIAA1566, PRKWNK3), belonging to the WNK family of kinases.
- It is a key component of the WNK3-SPAK/OSR1 kinase cascade that regulates electrolyte homeostasis and cell volume recovery during hyperosmotic stress. Related references: PMID:16275911, PMID:16275913, PMID:16501604
- Upon cell shrinkage, WNK3 acts as a molecular crowding sensor and undergoes liquid-liquid phase separation to form membraneless condensates that concentrate its substrates. Related references: PMID:36318922
