Biological Background: WNK1 Function and Localization

  • WNK1 (Lysine-deficient protein kinase 1) is a serine/threonine-protein kinase that functions as the master regulator of the WNK-SPAK/OSR1 kinase cascade, critically controlling ion cotransporter activity and systemic blood pressure (PubMed:15883153, PubMed:17190791, PubMed:31656913).
    Related references: PMID:15883153, PMID:17190791, PMID:31656913
  • Under hyperosmotic stress, WNK1 acts as a molecular crowding sensor, undergoing liquid-liquid phase separation to form membraneless compartments that concentrate WNK1 with its substrates OXSR1/OSR1 and STK39/SPAK, thereby enhancing downstream phosphorylation and regulatory volume increase (PubMed:36318922).
    Related references: PMID:36318922
  • Beyond its canonical kinase cascade, WNK1 exerts kinase-dependent functions in angiogenesis by activating OXSR1/OSR1 for chemotaxis and STK39/SPAK for endothelial cell proliferation (PubMed:25362046), and phosphorylates additional substrates such as SYT2, PCF11, and NEDD4L to regulate synaptic vesicle binding, mRNA export (PubMed:29196535), and membrane protein stability, respectively.
    Related references: PMID:25362046, PMID:29196535
  • A kinase-defective kidney-specific isoform acts as a dominant-negative regulator of the full-length WNK1, modulating sodium and potassium balance in the distal nephron, while the kinase-independent scaffold functions of WNK1 control membrane localization of various transporters and channels such as SLC4A4, SLC26A6, TRPV4, and CFTR, and promote SGK1 activation via mTORC2 recruitment (PubMed:14645531, PubMed:36373794).
    Related references: PMID:14645531, PMID:36373794
  • WNK1 is widely expressed with highest levels in testis, heart, kidney, and skeletal muscle, and shows specific presence in dorsal root ganglia and the distal/connecting tubules of the kidney, consistent with its roles in nociception and electrolyte homeostasis.
  • Subcellularly, WNK1 localizes to the cytoplasm, nucleus, and mitotic spindle, and participates in actin cytoskeletal reorganization, mitotic abscission, and negative regulation of autophagy (PubMed:10660600, PubMed:21220314, PubMed:27911840).
    Related references: PMID:10660600, PMID:21220314, PMID:27911840
  • Post-translational modifications include phosphorylation, ubiquitination, and glycosylation; the protein is also chloride-sensitive, with mutations linked to hereditary sensory and autonomic neuropathy type II (HSAN2) and pseudohypoaldosteronism.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-210) of human WNK1, which may recognize both full-length and kidney-specific isoforms—validate detection in relevant cell or tissue lysates.
  • For Western blot, a band near 250 kDa is expected; however, multiple isoforms and potential cleavage products may be observed. Consider using fresh protease inhibitors and optimized transfer conditions for high-molecular-weight proteins.
  • ELISA applications may benefit from using the purified recombinant protein or peptide as a positive control to establish assay sensitivity and specificity.
  • Cross-reactivity with mouse and rat has been reported; verify reactivity in your species of interest with appropriate controls (e.g., knockout lysates or blocking peptide) where possible.
  • When studying phosphorylation-dependent signaling, note that this polyclonal antibody is not phospho-specific; pair with phospho-specific antibodies to monitor WNK1 activation status.
