Biological Background: WNK1 Function and Localization

  • WNK1 is a serine/threonine-protein kinase that serves as a critical component of the WNK1-SPAK/OSR1 kinase cascade, regulating blood pressure and ion homeostasis. Under hyperosmotic stress, it acts as a molecular crowding sensor, undergoing liquid-liquid phase separation to create a membraneless compartment that concentrates WNK1 with its substrates OXSR1/OSR1 and STK39/SPAK, promoting their phosphorylation and activation.   • Activated OXSR1/OSR1 and STK39/SPAK phosphorylate and activate Na-K-Cl cotransporters (SLC12A1/NKCC2, SLC12A2/NKCC1) while inhibiting K-Cl cotransporters (SLC12A5/KCC2, SLC12A6/KCC3), thereby promoting ion influx and regulatory volume increase.   • For Western blot, note the large predicted molecular weight (~251 kDa); use a low-percentage SDS-PAGE gel (e.g., 6%) and wet transfer with extended time to ensure efficient transfer.
  • In immunofluorescence/immunocytochemistry, WNK1 localizes to the cytoplasm, nucleus, and mitotic spindle; appropriate fixation and permeabilization should be validated for the specific cell type and compartment of interest.
  • As a polyclonal antibody, lot-to-lot variability may occur; include positive (e.g., HeLa, HEK293) and negative controls in each experiment to confirm reactivity.
