Biological Background: WNK1 Function and Localization

  • WNK1 (Serine/threonine-protein kinase WNK1) is the core kinase of the WNK1-SPAK/OSR1 cascade, which is a critical regulator of blood pressure and cell volume; it promotes regulatory volume increase by sensing hyperosmotic stress and activating ion influx pathways.   • Under hyperosmotic conditions, WNK1 functions as a molecular crowding sensor, undergoing liquid-liquid phase separation to form membraneless compartments that concentrate WNK1 with its substrates OXSR1/OSR1 and STK39/SPAK, enhancing kinase signaling. Related references: PMID:36318922
  • Downstream, WNK1 phosphorylates and activates OXSR1/OSR1 and STK39/SPAK, which in turn phosphorylate ion cotransporters (SLC12A1/NKCC2, SLC12A2/NKCC1, SLC12A5/KCC2, SLC12A6/KCC3) to regulate sodium, potassium, and chloride transport, thereby controlling cell volume and blood pressure.
