Biological Background: WIPI2 Function and Localization
- WIPI2 (WD repeat domain phosphoinositide-interacting protein 2), also known as WIPI49-like protein 2, is encoded by the WIPI2 gene and belongs to the WIPI family of autophagy-related proteins.
- It acts as a core component of the autophagy machinery, regulating the formation of autophagosomes for lysosomal degradation. Related references: PMID:20505359 PMID:28561066
- WIPI2 functions in the early steps of preautophagosomal structure formation by binding phosphatidylinositol 3-phosphate (PtdIns3P) on endoplasmic reticulum membranes and mediating ER-isolation membrane contacts. Related references: PMID:28561066 PMID:28890335
- Upon activation, WIPI2 recruits the ATG12-ATG5-ATG16L1 complex to phagophore assembly sites, which is essential for ATG8 family protein lipidation and starvation-induced autophagy. Related references: PMID:20505359 PMID:28561066
- A specific isoform (isoform 4) is additionally required for xenophagy, binding to Salmonella-containing membranes and recruiting the ATG12-ATG5-ATG16L1 complex to clear pathogenic bacteria.
- The protein localizes to the membrane of preautophagosomal structures.
- WIPI2 is ubiquitously expressed, with high levels in heart, skeletal muscle, and pancreas; its expression is frequently downregulated in pancreatic and kidney tumors.
- Keywords associated with WIPI2 include alternative splicing, autophagy, disease variant (intellectual disability), lipid-binding, phosphoprotein, and WD repeat.
Experimental Guidance and Technical Tips
- The immunogen consists of a recombinant protein spanning amino acids 187-436 of human WIPI2, covering a region that may recognize multiple isoforms; consider verifying isoform specificity if needed.
- For Western blotting, a band around 49 kDa is expected; validate using positive control lysates from heart, skeletal muscle, or pancreas.
- For ELISA, a recombinant WIPI2 protein can serve as a standard; ensure appropriate blocking and incubation conditions.
- As with any polyclonal antibody, batch-to-batch variability may occur; always validate the antibody in your specific experimental system.
