Biological Background: WIPI1 Function and Localization
- WIPI1 is a WD repeat protein that functions as a key component of the autophagy machinery, mediating the sequestration of cytoplasmic cargo into autophagosomes for lysosomal degradation.
Related references: PMID:15602573, PMID:20114074, PMID:20484055
- It plays a critical role in starvation- and calcium-induced autophagy and in mitophagy, and it binds phosphatidylinositol 3-phosphate (PtdIns3P) to be recruited to phagophore assembly sites on the endoplasmic reticulum.
Related references: PMID:28561066, PMID:31271352, PMID:33499712
- Together with WIPI2, WIPI1 helps recruit the ATG12-ATG5-ATG16L1 complex, which is required for autophagosomal membrane elongation.
Related references: PMID:28561066
- It also cooperates with WDR45/WIPI4 to facilitate ATG2-mediated lipid transfer at PI3P-positive membranes.
Related references: PMID:31271352
- WIPI1 is involved in the xenophagic elimination of intracellular Staphylococcus aureus.
Related references: PMID:22829830
- In addition to autophagy, WIPI1 regulates melanogenic enzyme transcription and melanosome maturation, and it may control mannose-6-phosphate receptor recycling.
Related references: PMID:21317285, PMID:15020712
-
It localizes to the Golgi apparatus, endosomes, clathrin-coated vesicles, preautophagosomal membranes, and the cytoskeleton.
-
WIPI1 is ubiquitously expressed, with high levels in skeletal muscle, heart, testis, pancreas, and placenta, and is upregulated in multiple tumor tissues.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal 1-100 amino acid region of human WIPI1, ensuring target specificity. Validate reactivity in the relevant sample system with appropriate controls (e.g., overexpression or knockdown).
- For Western blot, expect a band near 49 kDa. Confirm specificity using WIPI1-knockout or siRNA-silenced cell lysates.
- ELISA optimization may require testing different coating conditions and antibody titrations. Given cross-reactivity with mouse and rat, consider using this antibody for comparative studies in rodent models.
- Autophagy induction (e.g., starvation or rapamycin treatment) can upregulate WIPI1 levels, which may enhance detection sensitivity.
