Biological Background: Wolframin Function and Localization

  • Wolframin is encoded by the WFS1 gene (aliases: WFS, WFRS, WFSL, CTRCT41) and is a 100 kDa transmembrane glycoprotein.
  • It localizes primarily to the endoplasmic reticulum (ER) membrane and is also found in cytoplasmic and secretory vesicles.
  • Wolframin plays a key role in regulating cellular calcium homeostasis by modulating the filling state of the ER Ca²⁺ store (PubMed:16989814). Related references: PMID:16989814
  • It negatively regulates the ER stress response and promotes the stability of V-ATPase subunits ATP6V1A and ATP1B1 by preventing their degradation (PubMed:23035048). Related references: PMID:23035048
  • Mutations in WFS1 are associated with Wolfram syndrome, characterized by diabetes insipidus, diabetes mellitus, optic atrophy, and deafness, as well as non-syndromic deafness and cataracts.
  • The protein is expressed at high levels in heart, brain, placenta, lung, and pancreas, with weaker expression in liver, kidney, and skeletal muscle, and is also present in pancreatic islet and beta-cell insulinoma cells.
  • Post-translational modifications include acetylation, phosphorylation, and glycosylation.
