Biological Background: WFS1 Function and Localization
- WFS1 encodes wolframin, a transmembrane glycoprotein of the endoplasmic reticulum (ER).
- Regulates cellular calcium homeostasis by modulating the filling state of the ER Ca(2+) store. Related references: PMID:16989814
- Negatively regulates the ER stress response and maintains the stability of V-ATPase subunits ATP6V1A and ATP1B1 through a proteasome-independent mechanism. Related references: PMID:23035048
- Localizes primarily to the ER membrane and is also found in cytoplasmic secretory vesicles.
- Highly expressed in heart, brain, placenta, lung, and pancreas; lower expression in liver, kidney, and skeletal muscle.
- Post-translational modifications include acetylation and phosphorylation (Acetylation, Phosphoprotein).
- Mutations in WFS1 are associated with Wolfram syndrome, as well as isolated low-frequency sensorineural hearing loss and cataract.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 70-170 of human WFS1, a region conserved in rat, consistent with the observed cross-reactivity. Confirm reactivity in your sample type.
- In Western blot, expect a band around 100 kDa. Signal is strongest in tissues with high WFS1 expression, such as heart and brain.
- For immunofluorescence, permeabilization is required to detect the ER membrane-localized target. Use ER markers as colocalization controls.
- For ELISA, this recombinant monoclonal antibody may serve as a capture or detection reagent when paired with a non-competing antibody.
