Biological Background: WDR81 Function and Localization

  • WDR81 is a WD repeat-containing protein that acts as a negative regulator of the PI3 kinase complex on endosomal membranes through interaction with BECN1, modulating phosphatidylinositol 3-phosphate levels to control endosome fusion, recycling, and transport. Related references: PMID:26783301
  • It facilitates the delivery of specific cargos such as BST2/tetherin from early to late endosomes, indirectly promoting their lysosomal degradation. Related references: PMID:27126989
  • WDR81 participates in aggrephagy, the selective autophagic degradation of ubiquitinated protein aggregates, by regulating SQSTM1 interaction with ubiquitinated proteins and recruiting MAP1LC3C. Related references: PMID:28404643
  • It may also play a role in maintaining normal mitochondrial structure and organization.
  • The protein is widely expressed with highest levels in the brain, particularly in the cerebellum and corpus callosum.
