Biological Background: WDR77 Function and Localization

  • WDR77 (also known as p44/Mep50, MEP-50) is a non-catalytic component of the methylosome complex, which also includes PRMT5 and CLNS1A. This complex catalyzes the symmetric dimethylation of specific arginine residues in spliceosomal Sm proteins and histones, a modification critical for targeting Sm proteins to the survival of motor neurons (SMN) complex for snRNP assembly (PubMed:11756452).   • The methylosome complex also methylates Piwi proteins (PIWIL1, PIWIL2, PIWIL4), which is required for their interaction with Tudor domain-containing proteins and subsequent localization to the meiotic nuage (PubMed:23071334). Related references: PMID:23071334

  • WDR77 is localized to both the nucleus and the cytoplasm, consistent with its roles in snRNP assembly and potential transcription regulation.

  • The protein is highly expressed in heart, skeletal muscle, spleen, testis, uterus, prostate, and thymus. In testis, it is expressed in germ cells and Leydig cells, but not in peritubular myocytes or Sertoli cells.

  • WDR77 belongs to the WD repeat protein family and is subject to phosphorylation. Alternative splicing generates multiple isoforms.

  • The immunogen corresponds to a region (aa 153-243) of human WDR77. For experiments with mouse or rat samples, verify sequence homology in the immunogen region to anticipate cross-reactivity, though the antibody is reported to cross-react with these species.

  • For Western blot (WB), a band near 37 kDa is expected. Optimize loading controls and antibody dilution for your specific sample type.

  • In immunofluorescence/immunocytochemistry (IF/ICC), consider appropriate fixation and permeabilization methods, and include controls to confirm nuclear or cytoplasmic localization.

  • For ELISA, validate the antibody with your specific antigen coating and detection system.
