Biological Background: WDR77 Function and Localization

WDR77 (also known as p44/Mep50, MEP-50) is a non-catalytic component of the methylosome complex, which also includes PRMT5 and CLNS1A. This complex catalyzes the symmetric dimethylation of specific arginine residues in spliceosomal Sm proteins and histones, a modification critical for targeting Sm proteins to the survival of motor neurons (SMN) complex for snRNP assembly (PubMed:11756452). The methylosome complex also methylates Piwi proteins (PIWIL1, PIWIL2, PIWIL4), which is required for their interaction with Tudor domain-containing proteins and subsequent localization to the meiotic nuage (PubMed:23071334). Related references: PMID:23071334

WDR77 is localized to both the nucleus and the cytoplasm, consistent with its roles in snRNP assembly and potential transcription regulation.

The protein is highly expressed in heart, skeletal muscle, spleen, testis, uterus, prostate, and thymus. In testis, it is expressed in germ cells and Leydig cells, but not in peritubular myocytes or Sertoli cells.

WDR77 belongs to the WD repeat protein family and is subject to phosphorylation. Alternative splicing generates multiple isoforms.
