Biological Background: WDR11 Function and Localization

  • WD repeat-containing protein 11 (WDR11), also known as bromodomain and WD repeat-containing protein 2 (BRWD2) and WD repeat-containing protein 15 (WDR15), is encoded by the WDR11 gene.
  • WDR11 is involved in the Hedgehog (Hh) signaling pathway and is essential for normal ciliogenesis. It regulates proteolytic processing of GLI3 and cooperates with EMX1 to induce downstream Hh pathway gene expression and gonadotropin-releasing hormone production. Related references: PMID:29263200
  • The WDR11 complex facilitates the tethering of AP-1-derived vesicles and collaborates with TBC1D23 to enable golgin-mediated capture of these vesicles at the trans-Golgi network. Related references: PMID:29426865
  • The protein localizes to multiple subcellular compartments, including the cytoplasm, cytoskeleton (cilium basal body and axoneme), nucleus, cytoplasmic vesicles, and the trans-Golgi network.
  • WDR11 is ubiquitously expressed across human tissues.
  • Keywords associated with WDR11 include chromosomal rearrangement, cell projection, Golgi apparatus, nucleus, disease variants linked to hypogonadotropic hypogonadism, intellectual disability, and Kallmann syndrome, phosphoprotein, and proto-oncogene.

Experimental Guidance and Technical Tips

  • For Western blot, the predicted molecular weight is approximately 137 kDa; consider using a low-percentage polyacrylamide gel and optimized transfer conditions to ensure efficient blotting of high-molecular-weight proteins.
  • Validate the antibody in relevant human cell lines or tissue lysates known to express WDR11 to confirm specificity and sensitivity.
  • In ELISA applications, the polyclonal nature may provide robust detection; consider pairing with a matched capture antibody if developing a sandwich immunoassay.
  • Optimal working dilutions should be determined empirically by the end user for each specific assay.
