Biological Background: WDFY4 Function and Localization
- WDFY4 (WD repeat- and FYVE domain-containing protein 4) is also known as C10orf64 and encodes a large protein of approximately 354 kDa containing WD repeat domains and a FYVE domain.
- It plays a critical role in regulating cross-presentation of viral and tumor antigens in conventional type 1 dendritic cells (cDC1). Mechanistically, it acts near the plasma membrane and interacts with endosomal membranes to promote antigen trafficking from endosomes to the cytosol.
- Involved in B-cell survival through the regulation of autophagy, linking the adaptive immune response to cellular degradation pathways.
- Localizes to early endosomes and the endoplasmic reticulum, consistent with its role in endosomal antigen trafficking and membrane dynamics.
- Associated with alternative splicing, producing multiple isoforms that may have tissue-specific or functional diversity.
- Contains WD repeats, which are known to facilitate protein-protein interactions, and a FYVE domain that binds phosphatidylinositol 3-phosphate, targeting it to endosomal membranes.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (amino acids 1–160) of human WDFY4; this region may be conserved across isoforms, but isoform-specific expression should be considered during validation.
- For Western blot (WB), the full-length protein is approximately 354 kDa; use appropriate low-percentage SDS-PAGE gels, extended transfer times, and high-sensitivity detection methods to resolve and detect this high-molecular-weight protein.
- Validate antibody specificity in relevant human cell types known to express WDFY4, such as dendritic cells or B-cell lines, using positive and negative controls.
- For ELISA, optimize coating antigen concentration and antibody dilutions using the recombinant immunogen or native protein from human cell lysates.
- Note that this antibody is reported to react with human samples; cross-reactivity with other species has not been tested and should be determined experimentally if needed.
