Biological Background: WBSCR22 Function and Localization

WBSCR22 (also known as BUD23, MERM1, or Williams-Beuren syndrome chromosomal region 22 protein) is an S-adenosyl-L-methionine-dependent methyltransferase that specifically methylates the N7 position of a guanine in 18S rRNA. It requires the methyltransferase adapter protein TRM112 for full activity. Related references: PMID:25851604

It acts as a locus-specific coactivator for steroid receptors (glucocorticoid, mineralocorticoid, androgen, progesterone), facilitating transactivation. WBSCR22 is required for maintaining open chromatin at the TSC22D3/GILZ locus and for histone H3K79 dimethylation, although direct histone methyltransferase activity has not been observed in vitro. Protein levels are elevated in bronchial brushings and normal lung, with expression in immune cells (B cells, T cells, macrophages), and tend to be down-regulated in inflammatory lung diseases and neoplasia.

Experimental Guidance and Technical Tips
- For Western blot, the predicted molecular weight is ~32kDa. Validate with appropriate positive controls (e.g., heart, skeletal muscle, or kidney lysates) and negative controls.
- In immunofluorescence/immunocytochemistry, expect nucleoplasmic and cytoplasmic staining, potentially enriched in the perinuclear region. Co-staining with organelle markers can help confirm localization.
