Biological Background: WBSCR22 Function and Localization

  • WBSCR22 (also known as BUD23, MERM1, or Williams-Beuren syndrome chromosomal region 22 protein) is an S-adenosyl-L-methionine-dependent methyltransferase that specifically methylates the N7 position of a guanine in 18S rRNA. It requires the methyltransferase adapter protein TRM112 for full activity. Related references: PMID:25851604

  • Beyond its catalytic function, WBSCR22 plays a critical role in pre-rRNA processing and 40S ribosomal subunit biogenesis/export independently of its methyltransferase activity. Related references: PMID:25851604 PMID:24086612

  • It acts as a locus-specific coactivator for steroid receptors (glucocorticoid, mineralocorticoid, androgen, progesterone), facilitating transactivation. WBSCR22 is required for maintaining open chromatin at the TSC22D3/GILZ locus and for histone H3K79 dimethylation, although direct histone methyltransferase activity has not been observed in vitro.   • Subcellularly, WBSCR22 localizes to the nucleoplasm and cytoplasm, with enrichment in the perinuclear region.

  • Widely expressed across tissues, with high levels in heart, skeletal muscle, kidney, and lung. Protein levels are elevated in bronchial brushings and normal lung, with expression in immune cells (B cells, T cells, macrophages), and tend to be down-regulated in inflammatory lung diseases and neoplasia.

Experimental Guidance and Technical Tips
