Biological Background: WBP11 Function and Localization
- WW domain-binding protein 11, also known as Npw38-binding protein (NpwBP), SH3 domain-binding protein SNP70, and splicing factor that interacts with PQBP-1 and PP1.
- Activates pre-mRNA splicing and may inhibit protein phosphatase 1 (PP1) phosphatase activity.
- Localizes to both the nucleus and the cytoplasm.
- Ubiquitously expressed; highly expressed at protein level in heart, pancreas, kidney, skeletal muscle, placenta, and brain; weakly expressed in liver and lung.
- Post-translationally modified by acetylation, methylation, phosphorylation, isopeptide bond formation, and Ubl conjugation, indicating tight regulatory control.
- Involved in mRNA processing and splicing, as well as rRNA processing, according to UniProt keywords.
- Associated with disease variants, highlighting its potential clinical relevance.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 540–641 near the C-terminus of human WBP11. Consider validating detection of full-length protein and any relevant isoforms in your sample system.
- For Western blot, the predicted molecular weight is approximately 70 kDa. Optimize lysate preparation and antibody dilution for human, mouse, or rat tissues as cross-reactivity data indicate recognition across these species.
- IHC-P may require antigen retrieval and careful titration. Test on positive control tissues known to highly express WBP11 (e.g., heart, pancreas).
- This antibody is also reported for IF/ICC and ELISA, but optimal working concentrations should be determined empirically by the end user.
