Biological Background: WAPL Function and Localization
- WAPL (Wings apart-like protein homolog), also known as friend of EBNA2 and WAPL cohesin release factor, is a critical regulator of sister chromatid cohesion and chromosome segregation during mitosis.
- It negatively regulates cohesin association with chromatin, ensuring proper removal of cohesin from chromosome arms in prophase and promoting accurate sister chromatid resolution. Related references: PMID:26299517
- The protein couples DNA replication to sister chromatid cohesion, which is essential for faithful chromosome partitioning in both mitotic and meiotic cells, and also contributes to DNA repair processes.
- WAPL localizes predominantly to the nucleus and chromosomes, but is also detected in the cytoplasm.
- Isoform 1 is highly expressed in uterine cervix tumors, while isoform 2 is widely expressed with particularly high levels in skeletal muscle and heart.
- Post-translational modifications include acetylation and phosphorylation, which may regulate its activity and interactions.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (amino acids 1–100), which is conserved in human WAPL isoforms; consider using peptide competition assays to validate antibody specificity.
- For Western blot, the predicted molecular weight is approximately 133 kDa; use appropriate gel percentages and transfer conditions to ensure efficient detection of this relatively large protein.
- For immunofluorescence and immunoprecipitation, optimize fixation and lysis conditions in your experimental system, and include appropriate controls (e.g., isotype control, knockdown/knockout validation).
- Cross-reactivity with mouse is reported; verify performance in the relevant species and tissue context before large-scale studies.
