Biological Background: VPS54 Function and Localization

  • VPS54 (Vacuolar protein sorting-associated protein 54) is also known as hepatocellular carcinoma protein 8 (HCC8), tumor antigen SLP-8p, and PPP1R164; it is a component of the GARP (Golgi-associated retrograde protein) complex.
  • Acts as a component of the GARP complex that mediates retrograde transport from early and late endosomes to the trans-Golgi network (TGN).
  • The GARP complex is required for the cycling of mannose 6-phosphate receptors between the TGN and endosomes, which is essential for proper lysosomal sorting of acid hydrolases such as CTSD.
  • Within the GARP complex, VPS54 is required to tether the complex to the TGN; it is not involved in endocytic recycling. Related references: PMID:18367545 PMID:25799061
  • Subcellular localization: Golgi apparatus, trans-Golgi network membrane.
  • Post-translational modifications and features: phosphoprotein, contains coiled coil domains, and is subject to alternative splicing.
  • Predicted molecular weight is approximately 111 kDa (UniProt annotation: 110,589 Da).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 420–720, a region likely encompassing coiled-coil motifs important for complex assembly; consider using this epitope information when designing competitive assays or truncation studies.
  • For Western blotting, a dilution series may be required to detect endogenous VPS54 at the predicted molecular weight of 111 kDa; validate in the relevant sample system.
  • In immunofluorescence/ICC, fixation and permeabilization conditions should be optimized to preserve Golgi/TGN membrane localization; co-staining with TGN markers (e.g., TGN46) is recommended.
  • ELISA can be applied for quantitative detection; ensure recombinant protein or peptide controls match the immunogen region for accurate quantification.
  • Cross-reactivity with human and mouse has been confirmed; for other species, perform sequence alignment and preliminary testing.
