Biological Background: VPS54 Function and Localization
- VPS54 (Vacuolar protein sorting-associated protein 54) is also known as hepatocellular carcinoma protein 8 (HCC8), tumor antigen SLP-8p, and PPP1R164; it is a component of the GARP (Golgi-associated retrograde protein) complex.
- Acts as a component of the GARP complex that mediates retrograde transport from early and late endosomes to the trans-Golgi network (TGN).
- The GARP complex is required for the cycling of mannose 6-phosphate receptors between the TGN and endosomes, which is essential for proper lysosomal sorting of acid hydrolases such as CTSD.
- Within the GARP complex, VPS54 is required to tether the complex to the TGN; it is not involved in endocytic recycling. Related references: PMID:18367545 PMID:25799061
- Subcellular localization: Golgi apparatus, trans-Golgi network membrane.
- Post-translational modifications and features: phosphoprotein, contains coiled coil domains, and is subject to alternative splicing.
