Biological Background: VPS39 Function and Localization
- VPS39, also known as hVam6p, TLP, or VAM6, is a Vam6/Vps39-like protein that functions as a regulator of TGF-beta/activin signaling. It inhibits SMAD3-dependent transcription while activating SMAD2-dependent transcription by interfering with SMAD3/SMAD4 complex formation.
- As a core component of the HOPS endosomal tethering complex, VPS39 plays a critical role in vesicle-mediated protein trafficking to lysosomal compartments, including endocytic transport and autophagy. The HOPS complex is recruited to Rab7 on late endosomal membranes and is essential for fusion of endosomes and autophagosomes with lysosomes.
