Biological Background: VPS37A Function and Localization

  • VPS37A, also known as HCRP1 (hepatocellular carcinoma-related protein 1), is a component of the ESCRT-I complex, a key regulator of the vesicular trafficking pathway.
  • It functions in the sorting of ubiquitinated endocytic cargo into multivesicular bodies (MVBs), facilitating the degradation of transmembrane receptors and other proteins.
  • The protein may also play a role in cell growth and differentiation, consistent with its decreased expression in hepatomas (hepatocellular carcinomas).
  • Subcellularly, VPS37A is localized to the late endosome membrane and can also be found in the nucleus.
  • It is widely expressed across human tissues including heart, brain, placenta, liver, skeletal muscle, kidney, and pancreas, with notably higher abundance in the liver.
  • VPS37A is subject to alternative splicing and is classified as a phosphoprotein, indicating regulatory post-translational modifications.
  • Mutations in the VPS37A gene have been associated with hereditary spastic paraplegia (SPG53), a neurodegenerative disorder, and the protein is implicated in liver cancer, highlighting its biomedical significance.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 224-397) of human VPS37A; this design may contribute to the antibody’s cross-reactivity with mouse and rat homologs.
  • For Western blotting, a band at approximately 44 kDa is expected; however, due to alternative splicing and phosphorylation, multiple bands or shifts may be observed in some samples.
  • In IF/ICC experiments, staining consistent with late endosome and nuclear localization should be anticipated; co-staining with organelle markers is recommended for validation.
  • ELISA can be employed for antibody titer quantification or antigen detection; optimal coating and detection conditions should be determined empirically.
  • Given the decreased expression in hepatomas, comparative analysis between normal and tumor liver tissue samples may be of interest when using this antibody.
