Biological Background: VPS35 Function and Localization
- VPS35 is a core component of the retromer cargo-selective complex (CSC), which prevents lysosomal degradation of transmembrane cargo by mediating retrograde transport from endosomes to the trans-Golgi network and recycling to the plasma membrane. Related references: PMID:15078903, PMID:15247922, PMID:20164305
- It interacts with various retromer-associated complexes, including SNX-BAR, SNX3, and SNX27, to facilitate distinct trafficking pathways; for instance, SNX3-retromer specifically transports Wntless (WLS). Related references: PMID:30213940
- As a recruitment hub, VPS35 connects to the WASH complex and TBC1D5, which are required for endosomal protein sorting and recycling. Related references: PMID:22070227, PMID:24819384, PMID:24980502, PMID:28892079, PMID:20923837
- VPS35 localizes primarily to endosomal membranes, including early and late endosomes, and is also present in the cytoplasm.
- The protein is ubiquitously expressed with highest levels in brain, heart, skeletal muscle, placenta, and other tissues.
- Mutations in VPS35 are linked to Parkinson disease, and it undergoes phosphorylation, underscoring its significance in neurodegeneration.
- VPS35 is involved in host-virus interactions, mediating human papillomavirus trafficking from early endosomes to the Golgi apparatus.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 100–200 of human VPS35; this antibody may recognize epitopes in the N-terminal region, suitable for detecting full-length protein.
- Validate the antibody in your specific sample system; the predicted molecular weight is 92 kDa, but post-translational modifications may shift band position in Western blot.
- This antibody has been tested in Western blot and ELISA; for other applications such as immunoprecipitation or immunofluorescence, pilot optimization is recommended.
- Due to cross-reactivity with mouse and rat VPS35, it can facilitate comparative species studies in retromer function and endosomal sorting.
