Biological Background: VPS33B Function and Localization
- VPS33B (Vacuolar protein sorting-associated protein 33B) is a member of the Sec1/Munc18 (SM) protein family, encoded by the VPS33B gene, and is also known as KDIDAR or PFIC12.
- It plays a critical role in vesicle-mediated protein trafficking to lysosomal compartments and membrane docking/fusion reactions of late endosomes/lysosomes.
- Specifically, VPS33B is required for proper trafficking and targeting of lysyl hydroxylase 3 (LH3) to intracellular collagen, impacting collagen modification (PubMed:28017832). Related references: PMID:28017832
- VPS33B mediates phagolysosomal fusion in macrophages (PubMed:18474358). Related references: PMID:18474358
- In complex with VIPAS39, VPS33B is involved in endosomal maturation, the apical recycling pathway, and maintenance of apical-basolateral polarity in epithelial cells (PubMed:20190753). Related references: PMID:20190753
- This protein localizes to late endosomes, lysosomes, early endosomes, clathrin-coated vesicles, and recycling endosomes membranes.
- VPS33B is ubiquitously expressed with highest levels in testis and low expression in the lung, and is associated with diseases such as arthrogryposis, renal dysfunction, and cholestasis (ARC syndrome) based on its disease variant keywords.
- It undergoes post-translational modifications including acetylation and phosphorylation, and exists in alternatively spliced forms.
Experimental Guidance and Technical Tips
- The immunogen covers the C-terminal region (aa 348-617) of human VPS33B; consider its potential to detect both full-length and any isoforms containing this region.
- For Western blot, anticipate a band near ~71 kDa (predicted molecular weight); using testis or high-expressing cell line lysates as positive controls is advisable.
- In immunofluorescence/ICC experiments, anticipate endosomal/lysosomal localization; co-staining with compartment-specific markers (e.g., LAMP1, EEA1) may help confirm specificity.
- For ELISA, the antibody’s reactivity can be tested with recombinant VPS33B or cell lysates; validate in the relevant sample system.
