Biological Background: VPS16 Function and Localization
- VPS16 (vacuolar protein sorting-associated protein 16 homolog), also known as hVPS16 or DYT30, is a core component of the HOPS and CORVET endosomal tethering complexes. It plays a critical role in vesicle-mediated protein trafficking to lysosomal compartments, including the endocytic and autophagic pathways. Related references: PMID:11382755, PMID:23351085, PMID:24554770
- VPS16 is required for the fusion of endosomes and autophagosomes with lysosomes; this function depends on its association with VPS33A (but not VPS33B) and involves STX17 but not UVRAG. Related references: PMID:25783203, PMID:24554770
- The protein localizes to late endosome/lysosome membranes, early endosomes, clathrin-coated vesicles, and autophagosomes, consistent with its role in tethering and fusion events.
- VPS16 is ubiquitously expressed across human tissues, supporting its fundamental role in membrane trafficking.
- Mutations in VPS16 are associated with dystonia (DYT30). The protein undergoes alternative splicing and nitration, and is identified in proteomics reference datasets.
- The full-length human VPS16 protein (839 amino acids) has a predicted molecular mass of approximately 95 kDa.
Experimental Guidance and Technical Tips
- The immunogen spans the C‑terminal region (aa 540–839); the polyclonal antibody may recognize full‑length VPS16 and possibly isoforms containing this segment.
- For western blotting, a predominant band near 95 kDa is expected in human, mouse, and rat lysates. Consider using cell lines with moderate to high VPS16 expression (e.g., HeLa, HEK293) as positive controls.
- In ELISA, the antibody can be applied as a capture or detection reagent; optimize coating concentration and blocking conditions for your specific assay format.
- Cross‑reactivity has been confirmed for human, mouse, and rat. Detection in other species may be possible due to sequence conservation but should be validated experimentally.
