Biological Background: VPRBP (DDB1- and CUL4-associated factor 1) Function and Localization

  • The DCAF1 gene encodes VPRBP (also known as DDB1- and CUL4-associated factor 1, HIV-1 Vpr-binding protein, or serine/threonine-protein kinase VPRBP), a large protein with multiple isoforms (observed at 118, 168, and 169 kDa) due to alternative splicing. It belongs to the DCAF family, characterized by WD40 repeats and a chromo domain.
  • VPRBP functions as the substrate recognition subunit of the CUL4A-RBX1-DDB1 E3 ubiquitin ligase complex (DCX complex).
