Biological Background: VPRBP (DDB1- and CUL4-associated factor 1) Function and Localization
- The DCAF1 gene encodes VPRBP (also known as DDB1- and CUL4-associated factor 1, HIV-1 Vpr-binding protein, or serine/threonine-protein kinase VPRBP), a large protein with multiple isoforms (observed at 118, 168, and 169 kDa) due to alternative splicing. It belongs to the DCAF family, characterized by WD40 repeats and a chromo domain.
- VPRBP functions as the substrate recognition subunit of the CUL4A-RBX1-DDB1 E3 ubiquitin ligase complex (DCX complex). This complex mediates ubiquitination and proteasomal degradation of target proteins such as NF2 and methylated substrates, recognized via the chromo domain. Related references: PMID:23063525
- As part of the EDVP complex, VPRBP directs ubiquitination of TERT, leading to telomerase inhibition, and of CCP110, impacting centrosome biology. Related references: PMID:19287380 PMID:23362280 PMID:28242748 PMID:34259627
- VPRBP acts as an atypical serine/threonine kinase that specifically phosphorylates histone H2A at Thr-120 (H2AT120ph) in a nucleosomal context, repressing transcription of tumor suppressor genes and contributing to oncogenesis. Related references: PMID:24140421
- Through interaction with LLGL1 and LLGL2, VPRBP participates in JNK-mediated apoptosis during cell competition, influencing tissue homeostasis. Related references: PMID:20644714
- Subcellularly, VPRBP localizes to the cytoplasm, nucleus, and centrosomes (microtubule organizing centers), consistent with its roles in nuclear ubiquitination and cytoplasmic signaling.
- VPRBP is ubiquitously expressed, reflecting its fundamental functions in cell cycle control, chromatin regulation, and stress responses.
- In the context of viral infection, VPRBP is hijacked by HIV-1 Vpr and HIV-2 Vpx to redirect cellular E3 ligase activity, leading to host protein degradation and facilitation of viral replication.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 1058–1396 in the C-terminal region of VPRBP. Consider validating isoform detection and potential cross-reactivity with other DCAF family members.
- Western blotting may reveal multiple bands (118, 168, and 169 kDa) due to alternative splicing and post-translational modifications. Use appropriate protein markers and validate in the relevant cell or tissue lysate system.
- For ELISA, optimize coating antigen concentration and antibody titration empirically; the polyclonal nature may require stringent blocking to reduce background.
- Mouse cross-reactivity is reported; however, confirm specificity in the intended mouse sample type, especially given possible sequence variations.
