Biological Background: VISTA/B7-H5/GI24 Function and Localization
- VISTA (V-set immunoregulatory receptor), also designated B7-H5, GI24, PD-1H, or SISP1, is a transmembrane protein encoded by the VSIR (C10orf54) gene. It belongs to the immunoglobulin superfamily.
- Acts as an inhibitory immune checkpoint receptor that suppresses T-cell activation (PubMed:24691993). Related references: PMID:24691993
- Has been implicated in the regulation of MMP2 activity via MMP14 (PubMed:20666777), and may modulate BMP4 signaling in embryonic stem cells (By similarity). Related references: PMID:20666777
- Predominantly localizes at the plasma membrane and shows high expression in placenta, spleen, lung, and peripheral blood leukocytes; moderate levels in lymph node, bone marrow, and uterus.
- Abundantly present on myeloid lineage cells (CD11b⁺ monocytes, CD66b⁺ neutrophils) and at lower levels on CD4⁺ and CD8⁺ T cells and a subset of NK cells; protein expression is absent on B cells.
- Contains V-type immunoglobulin domain, a signal peptide, a transmembrane helix, and multiple post-translational modification sites including N‑glycosylation and phosphorylation.
