Biological Background: VIPAS39 Function and Localization

  • VIPAS39 (SPE39, VIPAR, VPS16B) is a spermatogenesis-defective protein 39 homolog encoded by the human gene VIPAS39 (formerly C14orf133). Multiple isoforms exist through alternative splicing.
  • In epithelial cells, VIPAS39 forms a complex with VPS33B, playing a critical role in apical RAB11A-dependent recycling endosome trafficking and maintaining apical-basolateral polarity. Related references: PMID:20190753
  • VIPAS39 is also implicated in lysosomal trafficking, potentially via interaction with the core HOPS complex in a discrete endosomal population, independent of VPS33B. Related references: PMID:19109425
  • By similarity, VIPAS39 has a proposed function in vesicular trafficking during spermatogenesis.
  • It may be involved in the transcriptional regulation of E-cadherin (by similarity).
  • Subcellular localization includes cytoplasm, cytoplasmic vesicles, early endosomes, recycling endosomes, and late endosomes, consistent with its trafficking roles.
  • VIPAS39 is a phosphoprotein and has been linked to protein transport, differentiation, spermatogenesis, and transcription regulation (UniProt keywords).

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant fusion protein covering amino acids 1–220 of human VIPAS39. This N-terminal region may allow detection of full-length VIPAS39 and possibly some isoforms.
  • For Western blotting (WB), the predicted molecular weight is approximately 57 kDa. Validate antibody specificity in appropriate cell or tissue lysates from human or mouse origin.
  • ELISA can be used for antibody titer determination or screening; optimal coating conditions and detection systems should be empirically determined.
  • Consider including appropriate controls (e.g., knockout or knockdown samples) to confirm specificity in WB.
  • Due to its role in endosomal trafficking, experimental conditions that affect endosomal compartments may alter VIPAS39 localization or abundance; plan fixation and permeabilization accordingly if used in immunocytochemistry (not validated by supplier).
