- Vimentin is a class-III intermediate filament protein encoded by the VIM gene, widely expressed in mesenchymal cells and certain other non-epithelial cell types.
- It forms filamentous networks that attach to the nucleus, endoplasmic reticulum, and mitochondria, contributing to cellular structural integrity and organelle positioning.
- Vimentin plays a critical role in cell directional movement, orientation, and organization of cell sheets, and is involved in Golgi complex polarization at the leading edge of migrating cells.
- It protects the polarity protein SCRIB from proteasomal degradation and facilitates its localization to intermediate filaments in a cell contact-dependent manner.
- Vimentin may promote axon outgrowth and motor fiber repair by recruiting DSP to outgrowth tips.
- Together with LARP6, vimentin stabilizes type I collagen mRNAs (CO1A1 and CO1A2), linking the cytoskeleton to extracellular matrix production.

- For Western blot, a band around 54 kDa is expected; consider using whole cell lysates from fibroblasts or mesenchymal cell lines as positive controls.
- In IHC-P, optimize antigen retrieval and antibody dilution; cross-reactivity with mouse and rat makes it suitable for comparative tissue studies.
