- Villin-1 (VIL, D2S1471) is an epithelial cell-specific Ca2+-regulated actin-modifying protein that modulates microvillar actin reorganization.
- It exhibits multiple actin-regulatory activities, including nucleation, bundling, capping, and severing, regulated by Ca2+, PIP2, and LPA binding.
- Binding to LPA enhances SRC-mediated phosphorylation and inhibits actin-modifying activities, while PIP2 inhibits capping/severing but promotes bundling.
- Villin-1 regulates epithelial cell morphology, migration, invasion, and apoptosis; it protects against DSS-induced apoptosis by preserving mitochondrial integrity.
- It enhances HGF-induced epithelial cell motility, chemotaxis, and wound repair.
- During S. flexneri infection, its actin-severing activity facilitates bacterial intracellular movement and dissemination.
