Biological Background: βIII-Tubulin (TUBB3) Function and Localization
- βIII-Tubulin (TUBB3) is a neuronal-specific class III beta-tubulin isotype that assembles into microtubules, the essential cytoskeletal filaments composed of alpha- and beta-tubulin heterodimers.
Related references: PMID:34996871, PMID:38305685, PMID:38609661 - TUBB3 plays a critical role in axon guidance and maintenance, directing proper growth cone dynamics in developing neurons.
Related references: PMID:20074521 - Binding of the guidance cue netrin-1 (NTN1) to its receptor UNC5C triggers dissociation of UNC5C from polymerized TUBB3 in microtubules, enhancing microtubule dynamics and mediating axonal repulsion.
Related references: PMID:28483977 - During dorsal root ganglion (DRG) development, TUBB3 contributes to the projection of sensory axons toward the spinal cord, a process regulated by netrin-1/UNC5C signaling.
Related references: PMID:28483977 - Subcellularly, TUBB3 localizes to the cytoplasm and enriches in dynamic cell protrusions such as growth cones, lamellipodia, and filopodia.
- TUBB3 expression is normally restricted to the central and peripheral nervous system, but it is significantly upregulated in most cancerous tissues, making it a cancer biomarker.
- The protein undergoes various post-translational modifications, including phosphorylation, and is subject to alternative splicing; disease-associated mutations in TUBB3 are linked to intellectual disability and congenital fibrosis of the extraocular muscles (CFEOM).
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 351–450, covering a unique C-terminal region of βIII-tubulin; this may provide isotype specificity against other beta-tubulin family members.
- For Western blotting, a single band near 50 kDa is expected; consider using knockout validation controls to confirm band identification.
- The antibody is applicable to immunofluorescence (IF-P) and immunohistochemistry (IHC-P); optimal fixation and permeabilization conditions should be empirically determined for each sample type, as microtubule antigens can be sensitive to over-fixation.
- For immunoprecipitation, the rabbit monoclonal format offers high affinity and low background; pre-clearing of lysates and gentle detergent conditions are recommended.
- Cross-reactivity with mouse allows the antibody to be used in common mouse models of neurobiology and cancer; validate in the relevant tissue or cell line system.
