Biological Background: VEGFR3/FLT4 Function and Localization
- Vascular endothelial growth factor receptor 3 (VEGFR3), also known as FLT4, is a tyrosine-protein kinase receptor that binds VEGFC and VEGFD.
- It plays an essential role in adult lymphangiogenesis and embryonic cardiovascular development, promoting endothelial cell proliferation, survival, and migration.
- Upon ligand binding, VEGFR3 activates downstream signaling pathways including MAPK/ERK, JUN, and AKT1, and phosphorylates SHC1 and PIK3R1.
- Alternative splicing generates isoforms; isoform 2 has a truncated C-terminus with reduced signaling, while secreted isoform 3 may act as a decoy receptor.
- Subcellularly, VEGFR3 is found at the cell membrane, in the cytoplasm, nucleus, and can be secreted.
- It is widely expressed, with protein detected in endothelial cells and tissues such as liver, muscle, thymus, placenta, lung, testis, ovary, prostate, heart, and kidney.
