Biological Background: VEGFR2/KDR/Flk-1/CD309 Function and Localization
- Vascular endothelial growth factor receptor 2 (VEGFR2), also known as fetal liver kinase 1 (FLK-1) or kinase NYK, is a tyrosine-protein kinase receptor encoded by the Kdr gene in mouse.
- It acts as a cell-surface receptor for VEGFA, VEGFC, and VEGFD, playing an essential role in the regulation of angiogenesis, vascular development, vascular permeability, and embryonic hematopoiesis.
- Upon ligand binding, VEGFR2 activates multiple signaling cascades including PLCG1, MAPK1/ERK2, MAPK3/ERK1, and AKT1 pathways, promoting endothelial cell proliferation, survival, migration, and differentiation.
- Isoforms lacking a transmembrane domain, such as isoform 2, may function as decoy receptors to negatively regulate VEGFA- and VEGFC-mediated lymphangiogenesis.
- Subcellular localization includes cell membrane, cytoplasm, nucleus, endoplasmic reticulum, cell junction, and early endosome; it can also be secreted.
- Expression is detected in endothelial cells, with high levels in adult heart, lung, kidney, brain, and skeletal muscle, and lower levels in most other adult tissues.
- Key post-translational modifications include phosphorylation and glycosylation, and the receptor contains immunoglobulin-like domains and a kinase domain.
Experimental Guidance and Technical Tips
- Since the immunogen corresponds to the extracellular domain (aa 20-762) of mouse VEGFR2, this antibody may recognize both full-length and shed/soluble forms of the receptor.
- For Western blot, a band at ~150 kDa is expected under reducing conditions; however, glycosylation may cause a slight shift. Use appropriate molecular weight markers and validate with known positive controls from mouse endothelial tissue or cell lines.
- For ELISA, consider using serial dilutions of the antibody and compare with a negative control to optimize signal-to-noise ratio. Confirm specificity with recombinant VEGFR2 protein if available.
- As a polyclonal antibody, experimental conditions such as dilution factors should be optimized for each lot and application. Cross-reactivity with related family members (e.g., FLT1, FLT4) cannot be excluded; use specific inhibitors or knockout samples for validation.
