Biological Background: VEGFR1 Function and Localization
- VEGFR1, encoded by the FLT1 gene (also known as FLT, FRT), is a receptor tyrosine kinase that serves as a high-affinity receptor for vascular endothelial growth factor A (VEGFA), as well as VEGFB and placental growth factor (PGF).
- It plays a crucial role in embryonic vasculature development, angiogenesis, endothelial cell survival, migration, macrophage function, and cancer cell invasion.
- The receptor is located on the cell membrane and can be internalized into endosomes; alternative splicing also generates secreted and cytoplasmic isoforms.
- Tissue-specific expression includes placenta (isoform 2), corneal epithelial cells and vascular smooth muscle cells (isoform 3), and wide expression in vascular endothelial cells and peripheral blood monocytes.
- Ligand binding triggers activation of downstream signaling cascades. VEGFR1 mediates phosphorylation of PIK3R1, MAPK1/ERK2, MAPK3/ERK1, AKT1 at 'Ser-473', SRC, YES1, and PTK2/FAK1. Related references: PMID:16685275

- The recombinant immunogen spans the C-terminal region (aa 1100-1180) of human VEGFR1, which is part of the intracellular kinase domain. This design may facilitate detection of multiple isoforms containing this region.
- For Western blot (WB), a band around 151 kDa is expected for the full-length receptor. Reduced migration or additional bands may reflect glycosylation or alternative splicing variants; validate in the relevant sample system.
- For immunofluorescence (IF/ICC), permeabilization of cells is required to access the intracellular epitope. Consider using appropriate controls to confirm specificity.
- ELISA applications can be optimized by determining the optimal coating concentration and using validated detection antibodies. Cross-reactivity with mouse and rat VEGFR1 suggests suitability for orthologous protein detection in these species.
