Biological Background: VEGFA Function and Localization
- VEGFA participates in the induction of key genes involved in the response to hypoxia and in the induction of angiogenesis, such as HIF1A. Related references: PMID:35455969
- VEGFA is a growth factor active in angiogenesis, vasculogenesis and endothelial cell growth. It induces endothelial cell proliferation, promotes cell migration, inhibits apoptosis and induces permeabilization of blood vessels. Related references: PMID:34530889
- VEGFA binds to the FLT1/VEGFR1 and KDR/VEGFR2 receptors, as well as NRP1/neuropilin-1. One isoform binds the KDR receptor but does not activate downstream signaling pathways and inhibits tumor growth.
- The protein localizes to multiple subcellular compartments, including the cytoplasm, nucleus, endoplasmic reticulum, Golgi apparatus, and is secreted into the extracellular space and matrix.
- VEGFA is a glycoprotein with heparin-binding affinity and contains disulfide bonds, which are essential for its structural integrity and function.
- VEGFA is widely expressed, with higher expression in pituitary tumors compared to the normal pituitary gland. Certain isoforms exhibit restricted expression patterns.

- For Western blot, the predicted molecular weight is approximately 27 kDa; however, glycosylation and potential isoform variations may result in additional bands. Use appropriate positive controls and molecular weight markers.
- In IHC-P and IF/ICC, antigen retrieval and permeabilization steps may be necessary to access VEGFA in its various subcellular locations. Optimize detection conditions for your tissue or cell type.
