Biological Background: VEGFA Function and Localization
- VEGFA (Vascular endothelial growth factor A) is a long-form growth factor that plays a central role in angiogenesis, vasculogenesis, and endothelial cell proliferation, migration, and survival. - VEGFA participates in the hypoxic response by inducing key genes such as HIF1A, and it protects cells from hypoxia-mediated cell death. Related references: PMID:35455969
- A specific isoform of VEGFA binds to KDR but fails to activate downstream signaling, thereby inhibiting angiogenesis and suppressing tumor growth.

- For western blotting, a band around 27 kDa is reported under reducing conditions, but molecular weight may shift due to glycosylation or isoform differences; loading controls and knockout/knockdown validation are advised.
- In immunofluorescence/immunocytochemistry, both secreted and intracellular VEGFA can be detected; optimize fixation and permeabilization protocols to preserve epitopes.
- For ELISA applications, the antibody can be considered for capture or detection in sandwich formats, but pairing with a compatible antibody and thorough validation is necessary.
