Biological Background: VEGFA Function and Localization
- Vascular endothelial growth factor A (VEGFA), also designated vascular permeability factor (VPF), is a member of the PDGF/VEGF growth factor family. Multiple isoforms are produced through alternative splicing, alternative initiation, and alternative promoter usage.
- It acts as a key regulator of angiogenesis and vasculogenesis, stimulating endothelial cell proliferation, migration, and survival while increasing vascular permeability (PubMed:34530889). Related references: PMID:34530889
- VEGFA binds to tyrosine kinase receptors FLT1/VEGFR1 and KDR/VEGFR2, as well as neuropilin-1 (NRP1). An isoform that binds KDR without activating downstream signaling inhibits angiogenesis and tumor growth.
- It participates in the cellular response to hypoxia by inducing the expression of HIF1A and other hypoxia-regulated genes, thereby protecting cells from hypoxia-induced apoptosis (PubMed:35455969). Related references: PMID:35455969
- The protein localizes to multiple compartments: secreted forms are found in the extracellular space and matrix, while intracellularly it is detected in the cytoplasm, nucleus, endoplasmic reticulum, and Golgi apparatus, reflecting its synthesis and secretion pathway.
- Widely expressed in many tissues, with elevated expression observed in pituitary tumors compared to the normal pituitary gland.
- Post-translational modifications include N-linked glycosylation and intramolecular disulfide bonds, which are critical for proper folding and receptor binding.
- Keywords associated with VEGFA: angiogenesis, growth factor, mitogen, heparin-binding, developmental protein, differentiation, glycoprotein, secreted, disulfide bond, alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 207–327 of human VEGFA (P15692). For Western blot, a band around 27 kDa may be observed; confirm in your experimental system.
- For immunohistochemistry (IHC-P), antigen retrieval and antibody titration should be optimized using VEGFA-positive tissue sections or cell pellets.
- The antibody cross-reacts with mouse and rat VEGFA; validate performance in the relevant species and application by including appropriate positive and negative controls.
- For ELISA, consider using recombinant VEGFA as a standard and testing serial dilutions to establish the linear detection range.
- As a mouse monoclonal antibody, it may require an anti-mouse secondary antibody for detection; ensure compatibility with the detection system.
