Biological Background: VEGF-D Function and Localization
- VEGF-D (Vascular endothelial growth factor D), also known as c-Fos-induced growth factor (FIGF), is a secreted glycoprotein belonging to the PDGF/VEGF growth factor family.
- It functions as a growth factor active in angiogenesis, lymphangiogenesis, and endothelial cell growth by stimulating proliferation and migration, and also affects blood vessel permeability.
- During embryogenesis, VEGF-D may participate in the formation of the venous and lymphatic vascular systems, and in adults it helps maintain differentiated lymphatic endothelium.
- VEGF-D exerts its effects by binding and activating the receptor tyrosine kinases VEGFR-2 (KDR/FLK1) and VEGFR-3 (FLT4).
- The protein is proteolytically processed at pairs of basic residues to generate mature forms.
- Subcellular location: VEGF-D is secreted into the extracellular space.
- Tissue specificity: Highly expressed in lung, heart, small intestine, and fetal lung, with lower expression in skeletal muscle, colon, and pancreas.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 20–220 of human VEGF-D, which spans the conserved VEGF homology domain (excluding the signal peptide). This design may yield antibodies that recognize both full-length and proteolytically processed forms.
- For Western blot (WB), the predicted molecular weight is approximately 40 kDa, but glycosylation and dimerization can result in bands at higher molecular weights. Consider using reducing and non-reducing conditions and validate with appropriate positive control lysates (e.g., lung or heart tissue).
- For ELISA applications, this polyclonal antibody can be used as a capture or detection reagent. It is advisable to validate the assay with recombinant human VEGF-D and native samples.
- As a rabbit polyclonal, it offers multi-epitope recognition, which may enhance sensitivity in detection assays but requires careful lot-to-lot consistency evaluation.
