Biological Background: VEGF-D Function and Localization
- Vascular endothelial growth factor D (VEGF-D), also known as c-Fos-induced growth factor (FIGF), is a secreted growth factor belonging to the VEGF family.
- Promotes angiogenesis, lymphangiogenesis, and endothelial cell proliferation and migration; also modulates vascular permeability.
- Exerts its effects by binding and activating the receptor tyrosine kinases VEGFR-2 (KDR/FLK1) and VEGFR-3 (FLT4).
- Involved in venous and lymphatic vascular system development during embryogenesis and maintenance of differentiated lymphatic endothelium in adults.
- Highly expressed in lung, heart, small intestine, and fetal lung; lower expression in skeletal muscle, colon, and pancreas.
- The protein is secreted, undergoes proteolytic cleavage at pairs of basic residues, and is glycosylated, with a molecular weight of approximately 40 kDa (precursor or processed forms).
- Contains disulfide bonds and a signal peptide; belongs to the PDGF/VEGF growth factor family.
Experimental Guidance and Technical Tips
- The immunogen corresponds to an internal region (aa 200-300) of human VEGF-D, which may detect both precursor and cleaved forms; sequence alignment with mouse and rat homologs should confirm cross-reactivity.
- For Western blotting, consider using a standard protocol with the expected band(s) around 40 kDa; note that VEGF-D is proteolytically processed, so multiple bands may be observed depending on sample type.
- For ELISA, the antibody can be used as a capture or detection reagent; validate pairing with another clone or polyclonal for sandwich ELISA.
- Due to high tissue specificity, validate expression in appropriate positive controls such as lung, heart, or small intestine lysates.
