Biological Background: VE-Cadherin Function and Localization
- VE-cadherin (Cadherin-5) is a calcium-dependent cell adhesion protein belonging to the cadherin family, also known as 7B4 antigen or CD144, encoded by the CDH5 gene in humans.
- It mediates homophilic cell-cell adhesion, contributing to the sorting of heterogeneous cell types. Related references: PMID:21269602
- It plays a key role in endothelial cell cohesion and intercellular junction organization.
- Through association with α-catenin, VE-cadherin links to the actin cytoskeleton, and acts as a junctional anchor for AMOTL2 and MAGI1 to couple actin fibers to endothelial cell junctions. Related references: PMID:10861224
- It collaborates with KRIT1 and PALS1 to establish and maintain endothelial cell polarity and vascular lumen formation, mediated by recruitment of the Par polarity complex and RAP1B. Related references: PMID:20332120

- Subcellularly, it localizes predominantly at cell junctions (adherens junctions), the cell membrane, and in the cytoplasm.

- For Western blot, expect a band near 88 kDa; use endothelial cell lysates (e.g., HUVEC) as positive control and validate with appropriate loading controls.
- For IHC-P, optimize antigen retrieval conditions; vascular endothelium serves as a suitable positive control tissue.
- In ELISA, this antibody can be used for capture or detection; sandwich assays benefit from pairing with a clone targeting a non-overlapping epitope.
