Biological Background: VE Cadherin Function and Localization
- VE Cadherin, also known as Cadherin-5, CD144, or 7B4 antigen, is a classical cadherin adhesion molecule encoded by the CDH5 gene.
- It functions as a calcium-dependent cell adhesion protein that mediates homophilic interactions, contributing to the sorting of heterogeneous cell types. Related references: PMID:21269602
- In endothelial cells, VE Cadherin associates with alpha-catenin to link the actin cytoskeleton to cell junctions, and anchors proteins such as AMOTL2 and MAGI1. Related references: PMID:10861224
- It collaborates with KRIT1 and PALS1 to regulate endothelial cell polarity and vascular lumen formation, involving the Par polarity complex and RAP1B. Related references: PMID:20332120
- Subcellularly, it is located at cell-cell adherens junctions, the cell membrane, and also found in the cytoplasm.
- Tissue-specific expression includes endothelial cells (at protein level) and the brain. Related references: PMID:27338829, PMID:2059658
- VE Cadherin positively regulates reorientation of actin stress fibers and endothelial cell reorientation in response to mechanical forces. Related references: PMID:25795300
- Post-translational modifications include phosphorylation and glycosylation; it contains calcium-binding repeats, undergoes alternative splicing, and is cleaved on pairs of basic residues.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 259-358 of human VE Cadherin, a region likely conserved in mouse, supporting the observed cross-reactivity.
- For Western blot (WB), consider the predicted molecular weight of ~88 kDa when selecting gel percentage and transfer conditions; validate with appropriate controls.
- ELISA applications may require optimization of coating and detection conditions for optimal signal-to-noise ratio.
- As a polyclonal antibody, lot-to-lot variation is possible; we recommend validating performance in your specific experimental system.
