Biological Background: VE Cadherin Function and Localization
- Cadherin-5, also known as VE-cadherin, 7B4 antigen, or CD144, encoded by the CDH5 gene, is a calcium-dependent cell adhesion protein.
- It mediates homophilic cell-cell interactions, contributing to the sorting of heterogeneous cell types (PubMed:21269602). Related references: PMID:21269602
- Plays a central role in endothelial cell biology by controlling the cohesion and organization of intercellular junctions; associates with alpha-catenin to link to the cytoskeleton (PubMed:10861224). Related references: PMID:10861224
- Works in concert with KRIT1 and PALS1 to establish and maintain correct endothelial cell polarity and vascular lumen, mediated by the Par polarity complex and RAP1B (PubMed:20332120). Related references: PMID:20332120

- For Western blot, consider using endothelial cell lysates (e.g., HUVEC) as positive control; the predicted molecular weight is approximately 88 kDa.
- In immunohistochemistry (IHC-P), verify staining specificity in vascular-rich tissues such as placenta or brain, and include appropriate negative controls.
