Biological Background: VDAC2 Function and Localization
- VDAC2 is a non-selective voltage-gated ion channel that mediates the transport of anions and cations across the mitochondrial outer membrane and plasma membrane. Related references: PMID:8420959
- The channel adopts an open conformation at zero mV and a closed conformation at both positive and negative potentials, with two distinct channel populations exhibiting different conductance and ion selectivity properties. Related references: PMID:8420959
- VDAC2 binds various lipids including ceramide, phosphatidylcholine, and sterols; ceramide binding specifically promotes mitochondrial outer membrane permeabilization (MOMP) and the apoptotic pathway. Related references: PMID:31015432
- It associates with the translocase of the outer mitochondrial membrane (TOM) complex and PINK1 kinase at depolarized mitochondria, stabilizing PINK1 and triggering mitophagy by recruiting the E3 ubiquitin ligase PRKN. Related references: PMID:40080546
- VDAC2 catalyzes the scrambling of phospholipids across the outer mitochondrial membrane, a function independent of its channel activity, translocating both anionic and zwitterionic phospholipids.
- Subcellularly, VDAC2 is located primarily in the mitochondrion outer membrane, with additional presence in the plasma membrane.
- Tissue-specifically, VDAC2 is expressed in erythrocytes at the protein level and is found in all tissues examined, reflecting a ubiquitous expression pattern. Related references: PMID:27641616 PMID:8420959
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein spanning the full-length human VDAC2 (amino acids 1-309). For western blot (WB), anticipate a band at approximately 32 kDa; validate in the relevant sample system.
- This antibody is validated for WB and ELISA. For ELISA, consider both direct and indirect formats, and optimize coating conditions as needed.
- Cross-reactivity is reported for human, mouse, and rat. Confirm reactivity in your target species and tissue type prior to large-scale experiments.
- As a polyclonal antibody, batch-to-batch consistency should be confirmed for quantitative studies.
