Biological Background: VDAC1 / Porin Function and Localization
- VDAC1, also known as voltage-dependent anion-selective channel protein 1, porin, outer mitochondrial membrane protein porin 1, plasmalemmal porin, porin 31HL, porin 31HM, or hVDAC1, is a member of the eukaryotic mitochondrial porin family.
- Functions as a non-selective voltage-gated ion channel that permits the diffusion of small hydrophilic molecules and mediates the transport of anions and cations across the mitochondrial outer membrane and plasma membrane; involved in cell volume regulation and apoptosis. Related references: PMID:10661876, PMID:11845315, PMID:18755977
- Exhibits voltage-dependent gating: open at low or zero membrane potential with weak anion selectivity, and closed at potentials above 30–40 mV with cation selectivity. Related references: PMID:18755977, PMID:8420959
- Interacts with key signaling molecules such as ceramide, phosphatidylcholine, cholesterol, and oxysterol. Related references: PMID:18755977, PMID:31015432
- Plays a dual role in mitochondrial quality control and apoptosis: downstream of PRKN and PINK1, polyubiquitination promotes mitophagy, while monoubiquitination limits mitochondrial calcium influx to prevent apoptosis; also participates in the permeability transition pore complex (PTPC) and mediates cytochrome c release. Related references: PMID:32047033, PMID:15033708, PMID:25296756, PMID:20230784
- Localizes to the mitochondrial outer membrane, cell membrane, and membrane rafts; expressed in heart, liver, skeletal muscle, and erythrocytes. Related references: PMID:8420959, PMID:27641616
- Subject to various post-translational modifications, including acetylation, phosphorylation, and ubiquitination, which modulate its function in apoptosis and mitophagy.
Experimental Guidance and Technical Tips
- The immunogen (full-length recombinant protein, aa 1-283) covers the entire sequence, thus the polyclonal antibody may recognize multiple epitopes, providing robust detection in denaturing and native conditions.
- For Western blot, the expected molecular weight is approximately 31 kDa; perform appropriate positive and negative controls using lysates from human, mouse, or rat tissues/cells.
- In immunohistochemistry (IHC-P), consider heat-induced antigen retrieval and optimize antibody dilution; mitochondrial enrichment or fractionation may enhance signal.
- For ELISA applications, the antibody can be used as a capture or detection reagent; pairing with other VDAC1 antibodies may be tested for sandwich assay development.
- VDAC1 is highly conserved across mammals; cross-reactivity with other species not listed may be possible but should be verified individually.
