Biological Background: VDAC1 / Porin Function and Localization
- VDAC1 (Voltage-dependent anion-selective channel 1), also known as porin, is a 31 kDa multi-pass transmembrane protein belonging to the eukaryotic mitochondrial porin family.
- It functions as a non-selective voltage-gated ion channel, mediating anion and cation transport across the mitochondrial outer membrane and plasma membrane, and plays roles in cell volume regulation and apoptosis. Related references: PMID:10661876, PMID:11845315, PMID:18755977
- The channel adopts an open conformation at low membrane potential (weak anion selectivity) and a closed conformation at potentials above 30-40 mV (cation-selective). Related references: PMID:18755977, PMID:8420959
- VDAC1 is a key component of the permeability transition pore complex (PTPC), facilitating mitochondrial cytochrome c release and triggering apoptosis. Related references: PMID:15033708, PMID:25296756, PMID:20230784
- It participates in PRKN/PINK1-mediated mitophagy regulation; polyubiquitination by PRKN promotes mitophagy, while monoubiquitination lowers mitochondrial calcium influx, inhibiting apoptosis. Related references: PMID:32047033
- VDAC1 binds various signaling molecules, including ceramide, phosphatidylcholine, cholesterol, and oxysterol. Related references: PMID:18755977, PMID:31015432
- Additionally, VDAC1 catalyzes phospholipid scrambling across the outer mitochondrial membrane, independent of its channel activity.
- The protein is predominantly expressed in heart, liver, skeletal muscle, and erythrocytes, with subcellular localization at the mitochondrial outer membrane, plasma membrane, and membrane rafts. Related references: PMID:27641616, PMID:8420959
Experimental Guidance and Technical Tips
- As a polyclonal antibody, consider including appropriate negative controls (e.g., pre-immune serum or knockout lysate) when performing Western blot or immunohistochemistry.
- For Western blot, a single band around 31 kDa is expected; pre-stain markers or positive control lysates from heart, liver, or skeletal muscle tissues can aid confirmation.
- In IHC-P, antigen retrieval methods (heat-induced or enzymatic) may enhance signal; validate staining in tissues with known high expression such as human heart or liver.
- This antibody has been validated for ELISA; end-users may explore sandwich ELISA pairing with a monoclonal capture antibody targeting a different VDAC1 epitope.
- Cross-reactivity with mouse and rat VDAC1 has been observed; validate detection in the relevant species and sample system before large-scale studies.
