Biological Background: VDAC 1/2/3 Function and Localization

  • VDAC1 (Voltage-Dependent Anion Channel 1) is also known as porin 1, plasmalemmal porin, and outer mitochondrial membrane protein porin 1. It is a member of the VDAC family, which includes VDAC2 and VDAC3, all recognized by this antibody.
  • It functions as a non-selective voltage-gated ion channel that mediates transport of anions and cations across the mitochondrial outer membrane and plasma membrane, playing critical roles in cell volume regulation and apoptosis (PubMed:10661876, PubMed:11845315, PubMed:18755977). Related references: PMID:10661876 PMID:11845315 PMID:18755977
  • The channel adopts an open conformation at low or zero membrane potential and a closed conformation at potentials above 30-40 mV, with the open state being weakly anion-selective and the closed state cation-selective (PubMed:18755977, PubMed:8420959). Related references: PMID:18755977 PMID:8420959
  • VDAC1 is localized to the mitochondrial outer membrane, cell membrane, and membrane rafts, consistent with its dual role in mitochondrial and plasma membrane transport.
  • Post-translational modifications include acetylation and ubiquitination. Polyubiquitination by PRKN promotes mitophagy, while monoubiquitination decreases mitochondrial calcium influx to inhibit apoptosis (PubMed:32047033). Related references: PMID:32047033
  • It binds various signaling molecules like ceramide, phosphatidylcholine, cholesterol, and oxysterol, modulating its activity and role in apoptosis (PubMed:18755977, PubMed:31015432). Related references: PMID:18755977 PMID:31015432
  • Tissue expression is observed in erythrocytes, heart, liver, and skeletal muscle (PubMed:8420959, PubMed:27641616). Related references: PMID:8420959 PMID:27641616
  • VDAC1 also catalyzes phospholipid scrambling across the outer mitochondrial membrane, independent of its channel activity, and is involved in mitochondrial permeability transition pore complex (PTPC) formation and cytochrome c release during apoptosis (PubMed:15033708, PubMed:25296756, PubMed:20230784). Related references: PMID:15033708 PMID:25296756 PMID:20230784

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from the N-terminal 1-100 amino acids of human VDAC1, a region conserved among VDAC1/2/3. Consider verifying sequence alignment if cross-reactivity to other species or isoforms is critical.
  • For Western blotting, a band at ~31 kDa corresponding to VDAC1/2/3 is expected. Validate the antibody in your relevant cell or tissue lysate under optimal conditions.
  • This antibody has been shown to work in immunofluorescence (IF-P), immunohistochemistry (IHC-P), and ELISA. Appropriate controls and antigen retrieval methods should be optimized for your specific samples.
