Biological Background: VCP Function and Localization

• VCP, also known as p97, TERA, or CDC48, is an ATPase belonging to the AAA (ATPases Associated with diverse cellular Activities) family, encoded by the VCP gene.
• Essential for the fragmentation and reassembly of the Golgi apparatus during mitosis, and involved in the formation of the transitional endoplasmic reticulum (tER).
• Forms a ternary complex with UFD1 and NPLOC4 to bind ubiquitinated proteins and facilitate their export from the ER to the cytoplasm for proteasomal degradation. Mediates the ERAD of CHRNA3 in cortical neurons via the STUB1-VCP-UBXN2A complex. Related references: PMID:26265139
• Recruited to double-strand breaks in an RNF8/RNF168-dependent manner and promotes TP53BP1 recruitment. Experimental Guidance and Technical Tips

• The immunogen corresponds to the C-terminal region (amino acids 647–806) of human VCP; the resulting polyclonal antibody may recognize multiple epitopes and could detect full-length VCP as well as C-terminal fragments. For WB, a band at ~89 kDa is expected, but validate in your sample system.
• Cross-reactivity is observed with human, mouse, and rat VCP. When using rodent samples, confirm reactivity and adjust conditions as necessary.
• For IHC-P and IF/ICC, optimize antigen retrieval and permeabilization conditions, as VCP is present in multiple subcellular compartments (cytoplasm, nucleus, ER).
• In ELISA applications, this antibody can be used for either direct detection or as a capture/detection pair component after validation.
• As a polyclonal reagent, lot-to-lot consistency may vary; establish appropriate controls for reproducibility.
