Biological Background: VCAM1 Function and Localization
- Vascular cell adhesion protein 1 (VCAM1), also known as CD106 or INCAM-100, is a cell adhesion glycoprotein encoded by the VCAM1 gene and exists in multiple isoforms due to alternative splicing.
- VCAM1 primarily mediates leukocyte adherence to the endothelium through interactions with integrins, regulating transendothelial migration during immune surveillance and inflammation. Related references: PMID:31310649 PMID:10209034 PMID:22970700
- It also acts as a quality-control checkpoint in bone marrow, providing a 'don't-eat-me' signal within MHC class-I presentation to prevent phagocytosis. Related references: PMID:35210567
- The protein localizes to the cell membrane and can be secreted as a soluble form, which may have distinct functional roles.
- VCAM1 is characteristically expressed on inflamed vascular endothelium and on macrophage-like and dendritic cells in both normal and inflamed tissues.
- The protein is heavily glycosylated, contains immunoglobulin domains and disulfide bonds, and is subject to ubiquitin-like conjugation, as indicated by UniProt keywords.
- The precursor protein has a calculated molecular weight of ~81 kDa; multiple observed bands (38, 71, 74, 81 kDa) in Western blotting may reflect isoform diversity, glycosylation, or proteolytic processing.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide mapping to amino acids 640-739 of human VCAM1 (UniProt P19320), a region likely conserved across human, mouse, and rat, supporting multi-species reactivity.
- For Western blotting (WB), typical observed bands range from 38 kDa to 81 kDa due to isoforms and modifications; consider using reducing conditions and appropriate loading controls.
- In immunofluorescence (IF-P) and immunohistochemistry (IHC-P), VCAM1 staining is expected on the plasma membrane and possibly in secretory vesicles; validate with tissue known to have inflamed endothelium.
- ELISA can be used to detect soluble VCAM1 in serum or culture supernatants; note that the antibody recognizes an intracellular peptide epitope, so sample denaturation may be required for capture formats.
