Biological Background: VCAM1 Function and Localization
- VCAM1 is an immunoglobulin superfamily cell adhesion molecule that exists in multiple isoforms due to alternative splicing, with the full-length form containing multiple immunoglobulin-like domains.
- VCAM1 mediates leukocyte adhesion to vascular endothelium by interacting with integrins, a key event in immune surveillance and inflammatory responses. Related references: PMID:31310649 PMID:10209034
- VCAM1 engagement during inflammation triggers calcium channel activation and RAC1 signaling, promoting leukocyte transendothelial migration. Related references: PMID:22970700
- In bone marrow, VCAM1 functions as a quality-control checkpoint for cellular entry by presenting a 'don't-eat-me' signal during MHC class-I presentation. Related references: PMID:35210567
- VCAM1 is primarily expressed on the cell surface and can be shed from the membrane as a secreted form.
- VCAM1 is highly expressed on inflamed vascular endothelium, as well as on macrophage-like and dendritic cells in both normal and inflamed tissues.
- VCAM1 is a glycoprotein containing multiple immunoglobulin-like domains stabilized by disulfide bonds, and is subject to ubiquitin-like conjugation (Ubl).
Experimental Guidance and Technical Tips
- For Western blotting (WB), the predicted molecular weight is ~81 kDa; however, due to glycosylation and potential shedding, multiple bands or a higher apparent mass may be observed. Validate under reducing and non-reducing conditions.
- For immunofluorescence (IF-P), consider testing both live-cell and fixed/permeabilized protocols, as VCAM1 is surface-exposed but also secreted. Endothelial cell lines stimulated with inflammatory cytokines (e.g., TNF-α) may enhance signal.
- For ELISA, this polyclonal antibody can be used for antigen capture or detection; optimize coating buffer and detection system for your specific assay format.
- Since this is a rabbit polyclonal antibody, lot-to-lot consistency may vary; we recommend pilot testing each new lot in your system.
