- It serves as a major regulator of leukocyte adhesion to the endothelium by interacting with integrins, facilitating transendothelial migration during inflammatory responses.
- Upon binding to its ligands on activated endothelial cells, CD106 triggers calcium channel activation and the small GTPase RAC1, promoting leukocyte extravasation.
- In the bone marrow, VCAM-1 acts as a quality-control checkpoint for entry, providing a 'don't-eat-me' signal in the context of major histocompatibility complex (MHC) class-I presentation.
- The protein is localized to the cell membrane and can also be secreted; it contains immunoglobulin domains, disulfide bonds, and is glycosylated.
- It is expressed on inflamed vascular endothelium, macrophage-like cells, and dendritic cells in both normal and inflamed tissues, with notable expression in the bone marrow.
