Biological Background: VAV3 Function and Localization
- VAV3 is a guanine nucleotide exchange factor (GEF) that activates Rho family GTPases, including RhoA, RhoG, and to a lesser extent Rac1, by binding to their nucleotide-free states.
- It plays a critical role in angiogenesis; recruitment by phosphorylated EPHA2 is essential for EFNA1-induced RAC1 activation and vascular endothelial cell migration and assembly.
- In osteoclasts, VAV3 cooperates with SYK tyrosine kinase to mediate signaling through integrin αvβ1 (ITAGV-ITGB1), a pathway essential for proper cytoskeleton organization and osteoclast function, acting via RAC1 but not RHO.
- VAV3 is required for proper wound healing, specifically for phagocytic cup formation during macrophage phagocytosis of apoptotic neutrophils.
- It contributes to integrin β2 (ITGB2)-mediated macrophage adhesion, with a lesser role in β3 (ITGB3)-mediated adhesion, but does not affect β1 (ITGB1)-mediated adhesion.
- Isoform 1 and isoform 3 are widely expressed, with isoform 1 more abundant in adrenal gland, bone marrow, spleen, fetal brain, and spinal cord; isoform 3 is detected at very low levels in these tissues.
- The protein contains a Dbl homology (DH) domain for GEF activity, a pleckstrin homology (PH) domain, zinc finger domains, SH2 and SH3 domains, and is regulated by phosphorylation.
