Biological Background: VAPA Function and Localization
- VAPA (Vesicle-associated membrane protein-associated protein A), also designated VAP-33 or VAP-A, is encoded by the VAPA gene in humans.
- As an ER-resident protein, VAPA establishes membrane contact sites between the ER and endosomes via interactions with FFAT motif-containing proteins like STARD3 and WDR44, facilitating cholesterol transfer and nascent protein export. Related references: PMID:33124732, PMID:32344433
- VAPA can be recruited to the plasma membrane by OSBPL3, where the complex modulates RRAS signaling and attenuates integrin beta-1 activation, and has been implicated in ER morphology regulation and vesicle trafficking. Related references: PMID:25447204, PMID:16143324, PMID:11511104, PMID:19289470
- Subcellularly, VAPA localizes to the endoplasmic reticulum membrane, cell membrane, tight junctions, and the nuclear membrane.
- It is ubiquitously expressed across tissues.
- The protein features a coiled-coil domain, a transmembrane helix, and undergoes acetylation and phosphorylation; it is also involved in host-virus interactions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (aa 137-227) of human VAPA, potentially allowing detection of multiple isoforms; validate in the relevant sample system.
- For Western blot, expect a band near 28 kDa; use appropriate positive and negative controls.
- In IHC-P, optimize antigen retrieval and antibody dilution for the tissue of interest.
- ELISA can be performed with recombinant VAPA as a standard; titration of coating antigen and antibody may be needed.
- The antibody cross-reacts with mouse VAPA; consider species-specific validation if using mouse-derived systems.
