Biological Background: β-TrCP/BTRC Function and Localization

  • F-box/WD repeat-containing protein 1A (also known as E3RSIkappaB, beta-TrCP) functions as a substrate recognition component of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complex, mediating ubiquitination and proteasomal degradation of target proteins by recognizing phosphorylated substrates. Related references: PMID:10066435, PMID:10497169, PMID:10644755

  • SCF(BTRC) mediates ubiquitination of CTNNB1 to participate in Wnt signaling and ubiquitinates NFKBIA, NFKBIB, and NFKBIE to regulate NF-kappa-B activation; it also targets the p105 form of NFKB1 for degradation. Related references: PMID:12077367, PMID:12820959, PMID:10835356, PMID:11158290, PMID:14673179

  • Ubiquitination of CEP68 by SCF(BTRC) is required for centriole separation during mitosis. Related references: PMID:25503564, PMID:25704143

  • SCF(BTRC) controls clock-dependent transcription by degrading phosphorylated PER1 and PER2. Related references: PMID:15917222

  • Acts as a negative regulator of smoothened signaling by mediating ubiquitination of GLI2 and GLI3, promoting their processing into transcription repressors. Related references: PMID:16705181, PMID:16371461

  • Mediates ubiquitination and degradation of additional substrates including REST, mTORC1 regulators (DEPTOR, TFE3, MITF), and UBR2 in T-cell receptor signaling. Related references: PMID:18354482, PMID:21258371, PMID:22017875, PMID:22017876, PMID:22017877, PMID:38225265

  • Localizes to both the cytoplasm and nucleus. Tissue expression is reported in epididymis at the protein level. Keywords include host-virus interaction, alternative splicing, WD repeats, and Wnt signaling pathway.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1–100) of human BTRC. Consider this epitope location when designing blocking or competition experiments.
  • For Western blot, the predicted molecular weight is 69 kDa. Include appropriate loading controls and validate band specificity in your sample system.
  • Phosphorylation-dependent interactions are central to BTRC function; using phosphatase inhibitors during sample preparation may help preserve physiologically relevant complexes.
  • In IHC-P, test antigen retrieval conditions and confirm staining with appropriate tissue controls.
